Reversal of multidrug resistance by IFN-α in bladder cancer cell lines

Abstract Purpose:To evaluate the reversal of multidrug resistance by-interferon (IFN-α) in bladder cancer cell lines.Methods:The effect of recombinant human IFN-α on the cytotoxicity of adriamycin(ADM) in human bladder cancer cell line BIU87 and its multidrug resistant counterpart BIU 87/ADM were studied by MTT assay,ADM accumulation in above cell lines were also detected by spectrofluometric analysis.Results:The results showed that IFN-α (500 IU/ml) increased the cytotoxicity of ADM and enhance the accumulation of ADM in BIU87/ADM cells,but (IFN-α) (500 IU/ml) had no effects on the cytotoxicity of ADM to BIU87 cells,the level of mdr1 mRNA was unchanged as assessed by dot blot analysis.Conclusions:IFN-α has the reversal effect on multidrug resistance in bladder cancer cell lines.
