提要 含银复方消毒剂为含硝酸银、过氧化物的白色透明液体，含银离子512 mg/L，pH值为2.29。以载体定量杀菌试验检测其杀灭微生物效果与稳定性。结果，用其加5倍水的稀释液（pH 2.77）对布片上大肠杆菌、金黄色葡萄球菌、白色念珠菌作用7 min，杀灭率均为100％。其原液存放54℃下14 d，杀菌效果无明显变化；但有机物存在使杀菌作用减弱。

EXAMINATION OF GERMICIDAL EFFICACY AND STABILITY OF COMPOUND SILVER-CONTAINING DISINFECTANT

Abstract he compound silver-containing disinfectant is a white transparent liquid containing silver nitrate and peroxide. It contains silver ion 512 mg/L and has pH value of 2.29. The carrier quantitative germicidal test was used to examine its germicidal efficacy and stability. The results indicated that its dilute solution (pH 2.77) formed by adding 5 times of water with a 7 minute contact time killed 100% of Escherichia coli, Staphylococcus aureus and Candida albicans on cloth strips. The germicidal efficacy of its stock solution did not change significantly after storing at 54℃ for 14 days, but presence of organic substance made its germicidal efficacy decrease.

含银复方消毒剂为硝酸银辅以过氧化物而成的白色透明液体，原液含银离子512 mg/L，pH值为2.29。在实验室，对其杀灭微生物效果及稳定性进行了试验观察。现将结果报告如下。
1 方法
1.1 试验菌片的制备
试验菌为大肠杆菌（8099）、金黄色葡萄球菌（ATCC 6538）、白色念珠菌（ATCC 10231）。用含1％蛋白胨的0.03 mol/L磷酸盐缓冲液（PBS）将其分别制成菌悬液（观察有机物影响时，该悬液中尚含小牛血清）。取0.02 ml菌悬液，滴染于1.0 cm×1.0 cm脱脂白平纹布片上，置37℃恒温箱中干燥30 min制成菌片（回收菌数为5×105～5×106cfu/片）。
1.2 中和剂选择试验
试验用金黄色葡萄球菌菌片进行。试验分8组：① 消毒剂+菌片；② （消毒剂+菌片）+中和剂；③ 中和剂+菌片；④ （消毒剂+中和剂）+菌片；⑤ PBS+菌片（正常菌片对照）；⑥ 无菌PBS对照；⑦ 无菌中和剂对照；⑧ 未接种菌的培养基对照。试验结果，当第6、7、8组不长菌，第3、4、5组组间菌数相差≤10％，第1组不长菌或菌数远低于第2组，第2组菌数≥100 cfu/片时，表明所用中和剂及其浓度适宜〔1, 2〕。
1.3 载体定量杀菌试验
试验时，将每1片菌片置于5 ml消毒液（阳性对照为PBS）中。作用至规定时间，取出菌片，放入含5 ml中和剂溶液的试管中。中和作用10 min，敲打试管80次，以洗脱菌片上菌并混匀。取洗脱液0.5 ml作活菌计数，计算杀灭率。
1.4 稳定性试验
随机取包装好的含银复方消毒剂样品，置54℃恒温箱内14 d。于放置前、后，分别取样，检测杀灭金黄色葡萄球菌效果。
2 结果
2.1 中和剂试验结果
试验表明，用含1.0％吐温80、0.1％卵磷脂、0.5％硫代硫酸钠的溶液中和经加5倍水稀释后的含银复方消毒剂溶液（pH 2.77），结果可符合所定的要求（表1）。
2.2 杀灭微生物效果
经检测，以加5倍水的含银复方消毒剂稀释液对大肠杆菌、金黄色葡萄球菌、白色念珠菌作用3 min，杀灭率均达99.9％以上；作用7 min，杀灭率均为100％（表2）。
2.3 有机物对杀菌效果的影响
染于布片上的金黄色葡萄球菌悬液中含25％和50％小牛血清时，以加5倍水的含银复方消毒剂稀释液达到杀灭率为100％的作用时间，需由不含小牛血清时的10 min分别延长至15 min与20 min（表3）。
2.4 稳定性
经检测，于54℃温箱保存14 d，含银复方消毒剂加5倍水的稀释液对金黄色葡萄球菌的杀灭率无明显变化（表4）。
