提要 复方消毒清洁剂为含烷基二甲基苄基氯化铵(1.6%)、伯辛醇、聚氧乙烯醚等的溶液,主要用于消毒地板。为了解其杀灭微生物效果,进行了悬液定量杀菌试验。结果,以其1∶300稀释液对大肠杆菌、金黄色葡萄球菌作用10 min,以其1∶160稀释液对白色念珠菌作用10 min,杀灭率均≥99.95%。以其1∶200稀释液擦拭地板作用5 min,对地板表面自然菌灭除率>90%。杀菌效果随菌悬液中小牛血清浓度增加而下降。该剂于54~57℃贮放14 d,杀菌作用无明显变化。

EXPERIMENTAL OBSERVATION ON GERMICIDAL EFFICACY OF COMPOUND DISINFECTING DETERGENT

Abstract The compound disinfecting detergent is a solution containing alkyl-dimethyl-benzyl-ammonium chloride (1.6%), primary-capryl alcohol, polyoxyethylene ether, etc. It is used mainly for disinfection of floor. In order to know its germicidal efficacy, suspension quantitative germicidal test was carried out. The results indicated that the killing rates of Escherichia coli and Staphylococcus aureus exposed to its 1∶300 dilute solution for 10 min and of Candida albicans exposed to its 1∶160 dilute solution for 10 min were ≥99.95%. Wiping the floor with its 1∶200 dilute solution for 5 min killed >90% of the natural bacteria on the surface of the floor. Its germicidal efficacy decreased with increasing the concentration of calf serum in the bacterial suspension and did not change significantly after storing at 54~57℃ for 14 days.

复方消毒清洁剂主要成分为烷基二甲基苄基氯化铵、伯辛醇、聚氧乙烯醚等,原液含1.6%季铵盐。主要用于消毒地板。对其杀灭微生物效果与稳定性进行了试验观察。现将结果报告于下。
1 方法
1.1 中和剂选择试验
试验菌为金黄色葡萄球菌(ATCC 6538)与白色念珠菌(ATCC 10231)。试验分组为:① 消毒剂+菌悬液;② (消毒剂+菌悬液)+中和剂;③ 中和剂+菌悬液;④ (消毒剂+中和剂)+菌悬液;⑤ 正常菌对照;⑥ 未接种菌的培养基对照。试验结果,以第6组不长菌,第3、4、5组组间菌数相差不超过10%,第1组不长菌或菌数远少于第2组,第2组菌数≥100 cfu/ml时,判定所选中和剂及其浓度适宜〔1〕。
1.2 悬液定量杀菌试验
试验菌为金黄色葡萄球菌、白色念珠菌、大肠杆菌(8099)。将菌悬液0.5 ml与消毒液(阳性对照组用磷酸盐缓冲液)4.5 ml混匀。作用至预定时间,吸0.5 ml菌药混合液加入盛有4.5 ml中和剂的试管中,混匀。中和作用10 min,作活菌计数,计算杀灭率〔1〕。
观察有机物影响时,向菌悬液中加小牛血清。
1.3 地板表面消毒效果检测
以该剂1∶200稀释液涂擦地板,留出相邻不涂擦的地板作对照。随后分别借助5 cm×5 cm规格板,用蘸中和剂的湿无菌棉拭涂抹采样。将采样后棉拭头剪入装有5 ml中和剂溶液的试管中。敲打试管80次,以洗脱棉拭上所采菌并混匀。取洗脱液(或其稀释液)0.5 ml接种平皿,倾注普通营养琼脂并混匀。于37℃培养48 h,计数菌落数,计算灭除率〔2〕。
2 结果
2.1 中和剂试验结果
经试验,用含1%卵磷脂、1%吐温80的溶液中和试验浓度消毒剂,均达到所定要求(表1)。
2.2 杀菌效果
试验表明,以该剂1∶300稀释液对大肠杆菌与金黄色葡萄球菌作用10 min,杀灭率分别为99.96%与99.95%;以其1∶160稀释液对白色念珠菌作用10 min,杀灭率达99.96%(表2)。
